旱地农田不同耕作系统的能量/碳平衡 总被引:5,自引:0,他引:5
摘要:加强农田土壤保持耕作管理,科学认识和调控农田耕作系统能流碳流,提高农业生态系统固碳减排能力,对于减缓农业对全球温室效应的贡献具有重要意义。本研究以北方半湿润偏旱区山西寿阳旱作春玉米土壤保持耕作试验研究为基础,利用田间定位观测数据、辅助能投入参数,土壤呼吸田间原位测定,以及农业生态系统能量/碳平衡分析及碳循环过程模拟方法,综合分析和比较不同耕作(CT传统、RT少耕和NT免耕)系统能量/碳平衡及能-碳关联影响。与CT比较,采用RT和NT措施下工业能耗CO2-C损失降低约4%—12%(相当11—35 kg CO2-C?hm-2?a-1)。在RT和NT系统下耗能系数可降低约6%—10%,能量生产效率可提高约7%—12%。2006—2007年由田间原位测定土壤呼吸CO2-C释放通量估算,在玉米休闲期(尤其是秋耕处理后),NT条件下土壤呼吸速率一般为最低(NT NT(2005380)>CT(1987375)。不同耕作下的玉米籽粒产量与生育期土壤呼吸通量趋势基本吻合,如2006-2007年玉米产量(kg?hm-2?a-1)平均为,RT(5614268)>NT(5533564)>CT(5487278)。玉米籽粒产量与生育期土壤呼吸通量之间呈密切相关(R2=0.88)。本研究结果得出,RT和NT对农田耕作系统的影响呈碳汇效应,且一般为NT >RT;而CT处理表现为碳源。RT和NT通过增加土壤碳投入是维持和提高土壤有机碳的有效途径。 相似文献

Stroke is among the most frequent causes of death and adult disability, especially in highly developed countries. However, treatment options to date are very limited. To meet the need for novel therapeutic approaches, experimental stroke research frequently employs rodent models of focal cerebral ischaemia. Most researchers use permanent or transient occlusion of the middle cerebral artery (MCA) in mice or rats.Proximal occlusion of the middle cerebral artery (MCA) via the intraluminal suture technique (so called filament or suture model) is probably the most frequently used model in experimental stroke research. The intraluminal MCAO model offers the advantage of inducing reproducible transient or permanent ischaemia of the MCA territory in a relatively non-invasive manner. Intraluminal approaches interrupt the blood flow of the entire territory of this artery. Filament occlusion thus arrests flow proximal to the lenticulo-striate arteries, which supply the basal ganglia. Filament occlusion of the MCA results in reproducible lesions in the cortex and striatum and can be either permanent or transient. In contrast, models inducing distal (to the branching of the lenticulo-striate arteries) MCA occlusion typically spare the striatum and primarily involve the neocortex. In addition these models do require craniectomy. In the model demonstrated in this article, a silicon coated filament is introduced into the common carotid artery and advanced along the internal carotid artery into the Circle of Willis, where it blocks the origin of the middle cerebral artery. In patients, occlusions of the middle cerebral artery are among the most common causes of ischaemic stroke. Since varying ischemic intervals can be chosen freely in this model depending on the time point of reperfusion, ischaemic lesions with varying degrees of severity can be produced. Reperfusion by removal of the occluding filament at least partially models the restoration of blood flow after spontaneous or therapeutic (tPA) lysis of a thromboembolic clot in humans.In this video we will present the basic technique as well as the major pitfalls and confounders which may limit the predictive value of this model. 相似文献

Brown bears have lost most of their range on the European continent. The remaining western populations are small, isolated and highly endangered. The Dinaric-Pindos brown bear population is the western-most stable population and the fourth largest in Europe. It has been recognized as a potential source for recolonization of populations whose survival is at risk. Indeed, several translocations of Dinaric bears to Italy, Austria and France have recently been made. Despite the importance of the Dinaric bear population, its genetic status remains poorly understood. Using tissue samples from 156 hunted or accidentally killed Dinaric bears in Croatia, this study analysed genetic diversity at 12 microsatellite loci, as well as population structure and past reductions in size. In addition, a subset of 59 samples was used to assess diversity of the mitochondrial DNA control region. The results indicate that Dinaric bears have high nuclear genetic diversity, as compared to other extant brown bear populations, despite genetic evidence of a bottleneck caused by past persecutions. However, haplotype diversity was low, probably as a result of male-biased dispersal and female philopatry. Not surprisingly, no evidence of population sub-structure was found using nuclear markers, as the bear habitat has remained continuous and the highway network has been built only recently. Management should focus on maintaining habitat connectivity and keeping the effective population size as large as possible. In addition, when removing individuals, care should be taken not to further deplete the population of rare haplotypes. A coordinated transboundary management of the entire Dinaric-Pindos brown bear population should be a priority for its long-term conservation. 相似文献

Nature conservation should ideally build on the scientific recommendations that are concluded from applied conservation research, as well as on monitoring schemes that evaluate the effectiveness of recommendations. We considered as a case study a system of six protected areas located in the Eastern Rhodopes mountains in the southern part of the European Green Belt (EGB). To investigate nature conservation effectiveness, we reviewed 196 articles from scientific journals and books, eight doctoral and master theses, and 39 scientific reports regarding the Greek (one protected area, 428 km2) and the Bulgarian (five protected areas, 904 km2) part of the study area. We extracted 743 conservation recommendations, and through questionnaires completed by 10 local experts, we found that 74% of the recommendations were familiar for the experts. In the Greek (GR) and the Bulgarian part (BG) only 52% and 16%, respectively, of the recommendations were implemented, and only 15% (GR) and 3.1% (BG) were implemented and evaluated regarding their effectiveness. According to the experts, the main reasons for non-implementation and non-evaluation were absence or incompetence of the responsible authorities. Some recommendations obtained a remarkable low rate of implementation, such as those regarding agriculture and livestock rearing practices (GR: 29%, BG: 16%) or mammal conservation (GR: 0%, BG: 16%). Some other recommendations obtained rather high rates at least for Greece, such as tourism and environmental education (GR: 57%, BG: 42%) and bird conservation (GR: 57%, BG: 11%). We found that researchers and conservation managers at both sides of the Greek-Bulgarian border face similar implementation problems, related often to the lack of political will for nature conservation and establishment of competent authorities. Can seed dispersal by human activity play a useful role for the conservation of European grasslands?

Objective: To review the recent research into human‐mediated dispersal (HMD) in the European rural landscape, and explore the potential positive aspect of HMD for grassland conservation, in contrast to it's common association with the spread of invasive species. Methods: A literature search was undertaken to identify HMD vectors in the rural landscape for discussion regarding dispersal potential past and present, implications for management, and the identification of future research needs. Results: Grazing animals are important propagule dispersers, but the reduced movement of livestock through the landscape has also meant a reduction in seeds dispersed in this way. Other, non‐standard human‐mediated dispersal vectors such as clothing and motor vehicles can also transport seeds of many species, and HMD vectors often transport seeds with a variety of dispersal specialisations. Recommendations: There should be a greater movement of grazing animals throughout the landscape, either within larger grazing areas or between existing grasslands. Where this is not possible, other, more directed dispersal of propagules from species‐rich communities to target sites should be considered. The potential of non‐standard HMD vectors to make a positive contribution to biodiversity should be considered, but more research into all types of HMD vectors is important if we are to fully understand their role in the dispersal of plant species in fragmented landscapes. 相似文献

The first step of many metabolomics studies is quenching, a technique vital for rapidly halting metabolism and ensuring that the metabolite profile remains unchanging during sample processing. The most widely used approach is to plunge the sample into prechilled cold methanol; however, this led to significant metabolite loss in Synecheococcus sp. PCC 7002. Here we describe our analysis of the impacts of cold methanol quenching on the model marine cyanobacterium Synechococcus sp. PCC 7002, as well as our brief investigation of alternative quenching methods. We tested several methods including cold methanol, cold saline, and two filtration approaches. Targeted central metabolites were extracted and metabolomic profiles were generated using liquid chromatography-tandem mass spectrometry (LC-MS/MS). The results indicate that cold methanol quenching induces dramatic metabolite leakage in Synechococcus, resulting in a majority of central metabolites being lost prior to extraction. Alternatively, usage of a chilled saline quenching solution mitigates metabolite leakage and improves sample recovery without sacrificing rapid quenching of cellular metabolism. Finally, we illustrate that metabolite leakage can be assessed, and subsequently accounted for, in order to determine absolute metabolite pool sizes; however, our results show that metabolite leakage is inconsistent across various metabolite pools and therefore must be determined for each individually measured metabolite. 相似文献

AbstractPhenolic compounds such as catechol and resorcinol are toxic and persistent pollutants in the aqueous environment. Detection procedures such as chromatographic and spectrophotometric methods are time-consuming and require sophisticated instruments with skilled manpower. Development of a simple, cost effective, portable and disposable paper based biosensor could be a better alternative to the conventional methods. The present study attempted to develop a paper based biosensor by immobilizing horseradish peroxidase enzyme to detect catechol and resorcinol in aqueous samples. Horseradish peroxidase catalyzes the oxidation of phenolic compounds to semiquinones, which on reaction with a chromogen, 3-methyl 2-benzothiazolinone hydrazine (MBTH) gives faint pink to red color depending on the compound and its concentration in the sample is the basis for biosensing application. Different methods of enzyme immobilization on filter paper like physical adsorption, covalent coupling, and polysaccharide entrapment were executed. The performance of the various enzyme immobilization methods was evaluated by analyzing the developed color intensity using ImageJ software. Entrapment technique is the most effective method of immobilizing enzyme on the filter paper that produces the highest color intensity with better stability. The visible limit of detection (LoD) was observed as 0.45?mM (50?mg/L) for catechol and 0.09?mM (10?mg/L) for resorcinol in aqueous samples. 相似文献

Sugar profile and hydroxymethylfurfural (HMF) of Saudi honey were examined through high-performance liquid chromatography (HPLC) system equipped with refractive index and diode array detectors. The work was designed to assess the quality of various types of blossom honey i.e. Sider (Ziziphus spina-christi), Dhuhyana (Acacia asak), Sumra (Acacia tortilis), Qatada (Acacia hamulosa), Dhurum (Lavandula dentata), multiflora with majra (Hypoestes forskaolii), multiflora with herbs, Keena (Eucalyptus spp.) produced in the southwestern areas of the kingdom. Hierarchical cluster analysis (HCA), principal cluster analysis (PCA), and similarity and difference indices (SDI) were also applied to examine the possible grouping based on the studied quality parameters. Four main sugars (two monosaccharides i.e. fructose and glucose, two disaccharides i.e. sucrose and maltose) and HMF were investigated . The average values of fructose and glucose were in the range 33.10%–44.77% and 26.68%–37.91%, respectively. The maltose was present in all types of honey and its mean values were in the range of 0.37%–2.97%, while sucrose was absent in six types of honey, 0.25% in one unifloral honey, and 3.25% in one multi-floral honey. HMF was not detected in seven types of honey but was below the limit of quantification (0.13 mg/kg) in one type of honey. PCA displayed the accumulative variance of 79.96% for the initial two PCs suggesting that honey samples were not well distinguished by their sugar profile. Based on the sucrose and HMF contents, it was concluded that all types of blossom honey from the Asir province were of the best quality in the kingdom and met the international quality parameters. 相似文献
